Triple mouse M Double glass window, no artificial dura Behaving in Simon's 2-mode detection task Visual expert 5/10/18 Objective at 13.7, slightly angled but should be ok. Should do better next time. PMT at 445 Some motion but seems fine. Task, starting visual trials only. Using normal LED "house lights" Imaging at surface1, rougly center of the window Shielding seems fine. Depth 250 um Laser 33% Continuous imaging Field is not great but see enough neurons to proceed. Mainly in the center. Scanners at 2.5V. Maybe subsequent data makes little sense. 17500 up 2 stopped at 174805 Behavior was not great but ok. Visual ~70%, audio at chance. Got worse towards the end. =============================================================== 5/15/18 depth: 170um angle: ~10 deg a little bit of light leak from visual stimuli, maybe some movement but hopefully ok Signal looks good. Many lick tuned neurons. Best one so far. Shielding seems fine. Unfolding from posteriorlateral to anteriormedial. Should be fine though. depth: 190um laser power 34% location is pretty close to where we imaged last time. signal looks pretty good - many neurons. Chasing Z drift: 5000 up 1 10000 up 2 some major noise at around 12000-13000. had to open the door to switch on ambient light. 15000 up 2 18000 up 2 21000 up 2 25000 up 1 29000 up 3 32000 up 2 36000 up 2 42000 up 2 49000 up 2 53000 up 2 62000 up 1 65000 down 2 80000 seems like drift is fairly stable now. Animal has donly only 150 trials though... squinting a lot. Pupil tracking will presumably be not ideal. Mostly running on vision only to keep the mouse working. Hopefully we'll get enough trials together. Behavior is pretty weak, ~66% vision, 50% audio. Only 250 performed trials. =============================================================== 5/17/18 angle: 10.5 deg depth: 175um laser: 35% Some movement, light from the stimulus is visible pretty clearly. Can't get it better in place right now. Lots of neurons. SNR is not great - maybe there is some ambient light leaking in somewhere. Field is good - roughly in the center of the window. PMTs at 0.455. Field looks good. Many active neurons. MUCH better SNR as in the AI93 mice. Shielding/movement are good. Location should match ALM, based on widefield data. More anteriorlateral to earlier recordings. No red structural channel in this recording. Signal is good enough to be able to use the green channel alone. Using vessel shadows as markers. 20000 up 5 27000 up 5 First 100 trials are vision only, 80% vision afterwards. Performance is pretty bad - close to chance in both conditions. 47000 up 5 56000 up 5 Switched to all audio after 375 performed trials. Stopped after 440 performed trials. Performance at chance. Signal is good though and should be stable. =============================================================== 8/24/18 laser: 45% location: MM. More posteriormedial in the window. Should match based on widefield imaging. Depth: 180um Signal looks good, many neurons. Mild light artefact but should be ok. Using 1.25% Dextran dye to label vessels. Switched 15% audio after 100 performed trials. Visual detection is pretty low at ~60%.Switched to all audio after 350 performed tials. Visual performance still at 60% - not great... Otherwise good recording, pretty stable, lots of neurons. Stopped after 420 performed trials.
