Triple mouse M Double glass window, no artificial dura Behaving in Simon's 2-mode detection task Visual expert 5/21/18 Objective at 10.8 PMT at 445 Task, starting audio trials only. Using normal LED "house lights" Imaging at vessel, rougly center of the window Shielding seems fine. Depth 180 um Laser 32% Continuous imaging, signal looks good. Much better than other mice so far. Motion is not that bad, shielding seems fine as well. Should be good. Scanners at 2.5V. First file is bad, use the second one. Should be a stable recording throughout. Some more drift towards the end. Corrected accordingly. Should be fine. There is a bunch of neurons that are very driven by him cleaning is paws. Might be useful for something :) Towards the end of the session we switched to audio only. after ~400 performed trials we switched to vision only and then back to audio at the very end. Behavior is not great ~65% audio, 50% vision. Lots of trials: 500+ performed trials. =========================== 5/21/18 Objective at 10.8 PMT at 445 Task, starting audio trials only. Using normal LED "house lights" Laser at 34% depth is 250um Imaging roughly in the center of the window. Maybe more posterior to the last time. Chasing Z drift: 6000 up 2 8000 up 1 12000 up 2 16000 up 1 18000 up 1 22000 up 2 28000 up 2 32000 up 2 40000 up 2 46000 up 1 50000 up 2 54000 up 2 56000 up 2 moved a bit deeper for a moment to check if x and z drift are associated. doesnt seem that way. seems like neurons are also drifting slightly downwards over time. 60000 up 2 63000 up 2 71000 up 5 73000 up 2 ... couldnt take notes for a while. had problems with xy drift and other stuff that I needed to focus on. 101000 up 2 108000 up 2 imaging is not very stable. seems to drift in all three dimensions. damn! some neurons are fairly constant. have less faith in the upper part of the field. 110000 up 2 113000 up 2 115000 up 3 120000 up 2 125000 up 2 127000 up 1 134000 up 2 138000 up 2 145000 up 2 154000 up 4 159000 down 10 seems like this is closer to the original imaging plane. This session will be quite drifty overall. =========================== 6/06/18 Objective at 10.8 PMT at 445 Using normal LED "house lights" Laser at 34% depth is 200um Imaging roughly in the anterior center of the window. Should match ALM coordinates. Shielding seems fine Had to restart behavior after ~20 trials because camera crashed the paradigm. Old paradigm is the one that started with visual trials. New paradigm started with audio only. 12000 up 3 20000 up 2 26000 down 3 32000 up 2 35000 up 2 41000 up 3 44000 up 2 52000 up 3 56000 down 3 60000 down 2 didnt take notes on drift for a while - was hard to stay in the same plane. current position is 154 um 118000 up 2 122000 up 2 tried to change the plane a couple of times but it seems fairly stable. currently at 150. Many neurons were active over a pretty wide range. Not sure if the objective actually moved as indicated when trying to change the plane... maybe that means the recording was pretty stable :) Behavior was good ~75% audio, 50% vision. Eye got occluded at some point so pupile tracking will presumably not very good. Hopefully good session overall. =========================== 06/08/18 Objective at 10.8 PMT at 445 Using normal LED "house lights" Laser at 32% depth is 200um Imaging roughly in the anterior center of the window. Very little drift - performance at ~76% for audio, 50% vision. Stopped after 500 performed trials.
