SNR is not great but should be good enough. Many neurons. Red channel has tdTomato tag. Seems to show axons/dendrites instead of cell bodies but good for structure. Laser power is 44%. Ran at 0.8/0.2 for audio and vision after ~130 performed trials. All audio before that. Switched to all vision after 410 performed trials. Stopped after 475 performed trials. Should be a good recording. Good data/behavior. Right place. Also seems to have less motion than the other mice. =========================== 7/28/18 Imaging depth is 180um. Location should match MM. Anteriormedial from large blood vessel. Maybe too posterior. Signal is not as good usual and the objective is angled. Medial part is higher by ~60um. Will try to adjust and hopefuly do better next time. Laserpower is 44%. Some neurons, low SNR. First file is stack of 40um in 2um steps. Starting at 160um downwards. 25000 up 5 80000 up 5 Animal seems pretty disengaged after 320 performed trials. Switched to all vision to get some more visual trials in. Performance is good. 70% audio, 54% vision. Stopped after 415 performed trials. Drift seemed fine - should be stable recording although brightness was increasing over time. Not 100% sure about the location because the vessel images were not ideal. Should be at the mark from the widefield but there is a small chance it ended up more posterior. Check later! Changed objective angle from 7.2 to 8.5 after the session. =========================== 7/29/18 Objective is still not angled enough. Will increase more after session. Mscan crashed when trying to do a stack. Restart. Laser at 45%. Depth is 200um. Location should match ALM - I think I ended up a bit too posterior. The large vessel in the field is presumably the one that is at the post. end of where I marked ALM on the map. Should go more anterior/medial next time. First file is stack of 40um, starting at 180um depth in 2um steps. See some cells but not very active overall. Starting with audio only and will switch to mixed after 100 trials. Changed objective angle to 9.2 Using gel from Aki today to see if change in SNR is different.Signal seems slightly dimmer than before but not dramatically different: Should be ok. Coordinates should match ALM. Pretty sure this is right. Depth is 190um. Laser power is 48%. First file is 40um stack with 2um steps. Starting at 170um. Starting with all audio trials - first trial is vision. Shielding is good. Not a lot of movement. 10000 up 5 60000 up 5 73000 up 5 Seems like SNR is changing with the new gel as well. Switched to all vision after 390 performed trials. Stopped after 450 performed trials. Audio detection is ~71%, vision ~50%. Good session overall.
