Anteriormedial from large blood vessel. Maybe too posterior. Signal is not as good usual and the objective is angled. Medial part is higher by ~60um. Will try to adjust and hopefuly do better next time. Laserpower is 44%. Some neurons, low SNR. First file is stack of 40um in 2um steps. Starting at 160um downwards. 25000 up 5 80000 up 5 Animal seems pretty disengaged after 320 performed trials. Switched to all vision to get some more visual trials in. Performance is good. 70% audio, 54% vision. Stopped after 415 performed trials. Drift seemed fine - should be stable recording although brightness was increasing over time. Not 100% sure about the location because the vessel images were not ideal. Should be at the mark from the widefield but there is a small chance it ended up more posterior. Check later! Changed objective angle from 7.2 to 8.5 after the session. =========================== 7/29/18 Objective is still not angled enough. Will increase more after session. Mscan crashed when trying to do a stack. Restart. Laser at 45%. Depth is 200um. Location should match ALM - I think I ended up a bit too posterior. The large vessel in the field is presumably the one that is at the post. end of where I marked ALM on the map. Should go more anterior/medial next time. First file is stack of 40um, starting at 180um depth in 2um steps. See some cells but not very active overall. Starting with audio only and will switch to mixed after 100 trials. Signal is not great but should be ok. Movement and shielding are fine. First file is stack with 20frameAvg. Starting at 150um in 2um steps until 190um. First ~40 trials are pretty bad behavior. Seems to stabilize after that. ALl audio until 125 performed trials. Switched to 90/10% audiovisual after that. All vision after 358 performed trials. Signal is not great, laser power at 62%. See some lick-modulated neurons though. Light shield is good. 9000 up 5 16000 up 5 Animal seems a bit stressed - perhaps becase it feels the laser. Will try to keep this session short. Eye tracking will be compromised towards the end. 55000 up 5 Location seems to be a bit to posteriorlateral. The bottom right part of the imaging should match ALM. Some lick-modulated cells there. Should try to go there next time. Switched to all vision after 315 performed trials. Stopped after 365 performed trials. Auditory performance at 75%. Vision at 51%. =========================== 8/09/18 Both PMTs at 0.455. Imaging depper in MM - depth is 350 um. Signal is not good. Laser power at 65%. Shielding is good. Fire alarm interrupted beginning of the session - animal seems ok though. First 30 trials are all visual. Switched to all auditory after that. Behavior seems ok though. Very low signal - see some dim activity but will be very few cells in the field. There is one brighter cell in the middle of the field, so we might get 1 neuron :) More cells are visible later in the session. 29000 up 5 Switched on audiovisual after 125 performed trials. 80% auditory, 20% visual. 36000 up 5 43000 up 10 50000 down 5 Stpopped after 505 performed trials. Animal behavior was pretty good: 75% audio, chance at vision. Many more cells towards the end. Hopefully we captured enough activity for the model to work out. Consider rejecting some of the early data in if there is too little activity. Trying to take a second Z-stack after imaging. Will need to wait a bit and have the animal just run in the behavior. Second stack is centered around 345um in the software - should be close to the first one. =========================== 8/11/18 Both PMTs at 0.455. Imaging depper in ALM - depth is 350 um. Signal is not good. Laser power at 65%. Location should match superficial position better in this one. Don't see very much. Won't be a good recording. First 100 trials are audio only, performance is not very good.Will hopefuly get a bit better later but consider not using this first part. Also, horrible signal compared to later! Set to 80% adio after that. 75000 up 15 Signal is notably better in the second half of the recording and can see more neurons now. Would be really useful to know why this is. Tried warming the gel a little bit today but now by a lot. Remember to to do this next time! Performance is good: 75% audio, 50% Stopped after 510 performed trials. Should go good session if you use only the later part! As before, I took a second stack after the recording was finished. Should have much better quality as the first one. Checked power output immediately after taking the animal out: 115mW at 65%. Signal quality change is presumably not because of increase in laser power...
